Biomanufacturing-enabled personalized theranostic drug delivery materials and implantable device construction
Personalized medicine calls for therapeutic systems that can respond to dynamic physiological conditions rather than rely on fixed treatment schedules. Our research focuses on biodegradable materials and implantable devices that integrate sensing, drug delivery, and therapeutic regulation into a single platform. A key strategy is to exploit the controllable degradation of biodegradable metals for active drug delivery. Magnesium-based valves and polymeric encapsulation structures were developed to achieve electrically or magnetically triggered drug release, while multi-valve architectures enabled programmable pulsatile and sequential delivery of therapeutic agents. This concept was further extended to biodegradable bioelectronic interfaces, where drug-loaded PEDOT sponges were integrated with magnesium-based electrodes to combine physiological signal recording with electrically controlled drug release. The resulting systems demonstrated coordinated sensing and treatment in neural and cardiac models. To reduce the dependence on wired connections, wireless resonant sensors were integrated with magnetically triggered drug delivery modules, enabling physiological monitoring and on-demand treatment within a single implantable platform. The system was demonstrated for monitoring abnormal respiratory rhythms and regulating disease-related physiological states in vivo. These studies establish a material–device framework for biodegradable and programmable theranostic systems. Additive manufacturing and biomanufacturing offer further opportunities to tailor material composition, microstructure, device architecture, and patient-specific geometries, providing a route toward personalized intelligent implantable devices.
